Blood Stem-Cell Transplant Regimen Reverses Sickle Cell Disease in Adults

 A modified blood adult stem-cell transplant regimen has effectively reversed sickle cell disease in 9 of 10 adults who had been severely affected by the disease, according to results of a National Institutes of Health study in the Dec. 10 issue of the New England Journal of Medicine. The trial was conducted at the NIH Clinical Center in Bethesda, Md., by NIH researchers at the National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK), the National Heart, Lung and Blood Institute (NHLBI), and the National Institute of Allergy and Infectious Diseases.

“This trial represents a major milestone in developing a therapy aimed at curing sickle cell disease,” said NIDDK Director Griffin P. Rodgers M.D., a co-author of the paper. “Our modified transplant regimen changes the equation for treating adult patients with severe disease in a safer, more effective way.”

Sickle cell disease is caused by an altered gene that produces abnormal hemoglobin, the protein in normal red blood cells that carries oxygen throughout the body. When affected red cells lose oxygen, they collapse into a sickle, or C, shape and become stiff and sticky. Clumps of these cells block blood flow and can cause severe pain, organ damage from lack of oxygen, and stroke. Anemia often develops in people with the disease because sickle cells die off quickly and bone marrow does not make new ones fast enough.

In trials by other investigators, nearly 200 children with severe sickle cell disease were cured with bone marrow transplants after undergoing a regimen in which their own marrow was completely destroyed with chemotherapy. That regimen, however, had proven too toxic for adults, who have years of accumulated organ damage from the disease and are less able to tolerate complete marrow transplantation.

In contrast to the established method in children, this adult trial sought to reduce toxicity by only partially replacing the bone marrow. The much longer lifespan of normal red blood cells, compared to sickle red blood cells, allows the healthy cells to outlast and completely replace the disease-causing cells.

To achieve this goal, the investigators used a low dose of radiation to the whole body and two drugs, alemtuzumab and sirolimus, to suppress the immune system. Alemtuzumab depletes immune cells, but does not adversely affect blood stem cells. Sirolimus does not block the activation of immune cells, but inhibits their proliferation, creating a balance that potentially helps prevent rejection of the new stem cells.

The radiation favorably conditions the bone marrow, where donor stem cells move in and begin producing new, healthy red blood cells. After a median two and one half years follow-up, all 10 recipients were alive and sickle cell disease was eliminated in nine.

“Our patients have had a remarkable change in their lives,” said John F. Tisdale, M.D., the trial’s principal investigator in the NIH Molecular and Clinical Hematology Branch. “They are no longer being admitted to the hospital for frequent pain crises, they have been able to stop chronic pain medications, go back to school and work, get married and have children. Given these results, our regimen will likely have broad application to other nonmalignant diseases and can be performed at most transplant centers.”

 Transplanted cells or tissue are known as grafts. To reduce the possibility of the immune system’s rejection of the graft or development of graft-versus-host disease, in which immune cells from the donor attack the recipient’s tissues, investigators tested the patient and the potential donor to determine if they are a good immunological match. This is called human leukocyte antigen (HLA) typing.

The investigators performed HLA typing on 112 people with severe sickle cell disease and 169 healthy siblings. Of these, 10 patient-sibling identical matches were found. Blood stem cells collected from the blood of healthy donors were then infused into their siblings, ages 16 to 45 years.

This relatively low toxicity regimen allowed patients to become tolerant to the donor immune cells and to achieve stable mixed donor chimerism. Chimerism is a condition in which an individual has two genetically distinct types of cells in the blood. This mixture of host and donor cells was sufficient to reverse sickle cell disease. In most patients the donor’s red blood cells completely replaced the recipient’s.

“Remarkably, the treatment did not result in graft-versus-host disease for any of the participants,” noted Susan B. Shurin, M.D., acting director of the NHLBI. GVHD is a common complication of stem cell transplantation and can lead to serious problems, such as rash, diarrhea and nausea, liver disease, or death. “We are continuing to explore better treatments with fewer side effects to help the millions of sickle cell patients worldwide. This is a very important study because it lessens the toxicity of a therapy known to be highly effective.”

 In the United States, approximately 80,000 people have sickle cell disease, found mainly in people of African ancestry. It occurs to a lesser extent in people of Hispanic, Middle Eastern, Asian and white ancestry. Worldwide, millions of people have sickle cell disease. The pain and complications associated with sickle cell disease can have a profound impact on patients’ quality of life, ability to work, and long-term health and well-being.

One of the main obstacles in treating a larger number of African-Americans with sickle cell disease is the relative lack of an available HLA-matched donor. Dr. Tisdale explained, “Most white Americans can easily find a matched donor in the unrelated bone marrow or cord blood registries; yet when we screened a number of the people in our trial who were without an HLA-matched sibling donor, we could not find a compatible unrelated donor.”

However, there may be a way beyond this health care disparity, Tisdale indicated. If participants in the current trial continue to do well with their transplants it may be possible to move to what he calls “haplo-transplantation,” or a half-match from a sibling, parent or child. “This would allow most people with sickle cell disease to be treated and enjoy a better quality of life,” he said.

New Ways To Treat Kidney Damage Using Adult Stem Cells

Scientists have discovered that it may be possible to repair a patient’s kidney using cells taken from their own body. They have shown for the first time that cells in bone marrow are capable of turning into kidney cells.

The breakthrough could lead to new ways to treat kidney damage caused by cancer and other diseases.

It could also mean that doctors may eventually be able to restore function to patients suffering from kidney failure, and may pave the way to new gene therapy for kidney diseases.

The work has been carried out by scientists at the Imperial Cancer Research Fund, Barts Hospital, the London Hospital and Imperial College School of Medicine.

Early development
They focused on cells from the bone marrow at the very earliest stage of their development.

Kidney facts:
An estimated 100,000 people in the UK suffer from some form of kidney disease, excluding cystitis and kidney stones.
There are approximately 32,000 kidney patients receiving treatment in the UK.
50% of these have had a kidney transplant and are considered to be actively having treatment because they still require regular anti-rejection medication.
During 1999, 1,742 kidney transplant operations were performed in the UK and Ireland.
However, 5,056 people were actively waiting for a suitable organ for transplant.
Currently about 25% of grafts fail after five years.
At this stage this stem cells have the potential to develop into white blood cells, red blood cells or another type of blood structure called a platelet.
Scientists have already shown that these cells can transform themselves into liver cells.

The new work shows that they can turn into kidney cells too.

Professor Nick Wright, head of Imperial Cancer Research Fund’s Histopathology Unit, said: “This discovery is very exciting and means we have new ways to treat kidney damage caused by cancer or other diseases.

“Doctors could use stem cells from the patient’s own bone marrow to replenish kidney cells lost by injury.

“This would be of huge benefit as the kidney is very poor at repairing itself.

“Furthermore, there would be much less complication with the kidneys rejecting the new cells, because they would come from the patient’s own body.

“Another exciting development would be using bone marrow stem cells containing genes resistant to cancer or other disease, to protect the kidneys from further damage.”


Research pathologists in Imperial Cancer’s Histopathology Unit analysed female kidneys transplanted into male patients.

Using a special DNA probe that identifies male cells they checked the patient’s new kidneys.

They found male kidney cells in the donated female kidneys, meaning that the recipient’s male bone marrow cells had transformed into kidney tissue.

Professor Wright said: “Anti-rejection medication after a kidney transplant costs about £5,000 per patient a year, and each year the number of new patients needing kidney transplants increases by about 5%.

“It would be fantastic to save kidney patients this trauma and save the NHS some money.”

Dr Poulsom, a research pathologist at Imperial Cancer, said: “The potential for advances in medicine from using adult stem cells is enormous.

“They can give rise to many different types of cells so any organ may one day be repaired. Using adult stem cells also avoids the ethical dilemmas associated with embryonic stem cell work.”

The National Kidney Research Fund said the research opened up exciting possibilities.

But in a statement, it said: “However, in spite of today’s announcement, the Fund believes that we are probably many years away from preventing kidney failure.

“This is because the intricate structure of the kidney compared to the simple liver tissue makes it much more difficult to replace the many functions of the two million kidney filters and tubules and also in kidney failure scarring distorts the tubules so that there is nowhere for new cells to function making it difficult for the kidney to repair itself.”

The research is published in the Journal of Pathology.

The Day Our Nation Stopped To Pray –
September 11, 2001

 Today we watched in horror as Lower Manhattan rocked.
      And we in this great nation stood and watched in shock.
Innocent victims on four separate planes had been flying here and there.
        They were taken hostage, to be killed. We wonder “Who would dare?”
The World Trade Center Twin Towers was the first target.
        At 8:45am, near the top, tower one took a direct hit.
Then with cameras rolling live before our eyes,
        A second flight, the same outcome, we watch these people die.
Tower Two is hit lower. The explosion is unbelievable.
        How could anyone hate so much? Is there really anyone that capable?
The madness is not over onto Washington and the Pentagon.
        Another plane, full of people, being used as a bomb.
Back to the Twin Towers – tower two begins to sway.
        A roar from the implosions as the buildings blown away
There are people jumping, falling metal and shards of glass.
        Ash and paper fill the air as billowing smoke goes rushing past.
Policemen and firemen those sent to render aid.
        Beneath the rubble of this tower is where their bodies lay.
Back to the Pentagon, flames are coming from the south wall.
        Calls coming in from the north end say they see smoke in the hall.  
From the pictures we can see a large and gaping hole.
        Fires being fought. Tears being shed. We wonder “Who is the foe?”
Back to tower one as it shudders beneath the weight.
        Cries for help, more people jump, for them help comes too late.
Again we watch in horror, as tower one implodes.
        How many were still inside? No one really knows.
Breaking news. We have confirmed a fourth flight has gone down.
        It crashed in Pennsylvania. Speculation – it was DC bound.
“Today our nation saw evil.” I hear President Bush say.
        What should we do? He suggests we pray.
Today we lost Americans to terrorists from afar.
        And the senselessness of their actions, will weight heavy on our hearts.
The Statue of Liberty flame burns bright, our Flag still flies today
        Today we are united. Today our nation stopped to pray.
As I get up from my knees and wipe away my tears.
        The questions come but answers don’t. Where do we go from here? 
I ask God for the answers, as I look to Him above.
         “Each and every one are safe. I surrounded them with Love”
“Angels were there with them, no one was left alone.
         They are gathered here with me. Now Heaven is there home.”
We sit and watch, trying to access, the events of this day.
        The day Americans came together. The day our nation stopped to pray.

Cancer Protocol Revised 2009

On December 23, 1971, the National Cancer Act was signed into law. Congress believed that a cure could be found by the Bicentennial in 1976. The war on cancer stalled before it got started. In 1989, one million Americans were diagnosed with a malignancy as well as over 500,000 new cases of skin cancer. Over 500,000 people died of cancer in America alone. The “New England Journal of Medicine” declared that the $30 Billion, 27 year war against cancer had been lost and admitted defeat for the entire field of conventional cancer treatment.

The American Cancer Society estimates 563,700 cancer deaths in 2004, that is more than 1500 people per day, and they state that the scientific evidence suggests that about one-third of those cancers will be related to nutrition, physical inactivity, obesity, and other lifestyle factors and could also be prevented. They estimate 180,000 cancer deaths due to tobacco use and another 19,000 deaths due to excessive alcohol use. It is time we start accepting a little personal responsibility for our actions, our choices, for ourselves.

The world turns upside down when you hear your doctor say “You have cancer.” or worse, “You have TERMINAL cancer.”. You experience a rush of emotions, thoughts and questions; “Why did this happen to me?”, “What will happen if …?”, “What can I do now?”. My mother experienced these emotions and asked these questions when she was diagnosed with terminal cancer – I needed answers. Thus began my journey. After eleven years of extensive reading, studying scientific research, medical journals, clinical trials, autobiographies of cancer survivors and numerous other publications covering both conventional and alternative cancer therapies, I have gained a comprehensive understanding of cancer, its effect on the body, how to restore balance and rebuild the immune system. Positive steps that allow the body to heal itself.

The first step is to realize that there are others, just like you, who have attacked cancer and won! Next, you should be asking yourself; “How did this start?” and “How do I win this battle?” The keys to winning the war against cancer are to educate yourself, using available research and resources, then make informed decisions for the best approach and treatment plan that fit your specific needs. These steps will empower you to take control of your illness. You must realize everything from here on out is YOUR DECISION. Finally, you must COMMIT to winning the war against the enemy within – your cancer. YOU HAVE THE MOST TO GAIN OR THE MOST TO LOSE – CHOOSE WISELY!


 I would wage a full-scale war on the cancer and fight like my life depends on it . . . because it does. I would take charge of my health. The doctors are there to consult, not to make decisions. Family doctors, oncologists, nutritionists, family and friends have opinions and advice regarding what to do, but the ultimate decision is mine . . . and it needs to be an informed decision, not an emotional one. See, no one can heal a person, or cure a disease. For example, urgent care physicians, when faced with a life threatening emergency, recognize the power of the body. They work to stabilize the patient’s body; then wait for the body to heal itself. Your challenge is no different – stabilize your body by eliminating the physical and psychological factors that contributed to your illness, fortify the body with nutritional elements that strenghten your body and encourage healing, focus your determination to fight the battle and your will to win! The three pronged attack involves your body, your mind and your spirit, working together with one purpose – restore your health. I can help people to learn how to take better care of themselves, help them learn how to nourish their body properly with diet and supplements, so they can promote and maintain health.

 I can tell you that I would not choose any therapy that would compromise my immune system. How can you expect to deal with poisons and chemical toxicity if your immune system is so weak that it is allowing cancer to thrive? If your path includes chemotherapy make sure your immune system is strong; so as to minimize the side effects. The side effects of these drugs can be worse for the body than the cancer and could actually cause death. If you have been at the bedside of a loved one through a conventional battle with cancer as I have, witnessed their body falling apart, toxic, bloated, bed ridden in pain – with Morphine as their only ally – then you would understand its effects.

 I would change my lifestyle. This diagnosis needs to be your wake-up call. Whether you are a single mother, a father trying to support your family or a CEO of a major corporation you must make the time to take care of yourself. Otherwise, the reality is you might not be here to worry about life’s other challenges. Imagine cancer as your worst houseguest. Now, imagine you invite them into your home. You give them your bedroom; you cook and clean for them; you wait on them hand and foot day in and day out. How hard do you think it will be to get them to leave? This is cancer! Your uninvited guest, the cancer, is less likely to leave your body if you are willing to be a good host. You must make a change.

Eliminate stress and everything negative from your life. Exercise is very important as it promotes oxygenation of the tissues, can lift your spirits and process fluids and toxins through the lymph nodes. Spending five minutes twice a day on a mini trampoline will help stimulate the lymphatic system. Taking a brisk walk while swinging your arms, swimming and aerobics or water aerobics are excellent. Also, do deep breathing exercises. Exercising for thirty minutes three times per week is a good place to start. Also, eliminate deodorants that contain aluminum.

I would stop drinking treated city water. I would drink 10 to 12 – 8 oz. glasses of filtered water per day to carry out the toxins. Minimum half your body weight in ounces of water each day. Increase during cleansing.

I would switch to a vegetarian, organic diet. Begin feeding your body, not the cancer. I would eliminate all meat and animal proteins, dairy, sugar (real or artificial sweeteners), bleached flour, salt, coffee, soda, tea (except herbal tea), corn syrup, hydrogenated oils, all processed foods, fried or fast foods and alcohol. I would eat a wide variety of raw and steamed vegetables including carrots, celery, asparagus, cucumber, squash, yams, pumpkin, and cruciferous and green leafy vegetables. Also include beets, parsley and cabbage as they aid in detoxification. I would eat a combination of fresh fruits including grapes, berries, apples, cherries, melon and limited amounts of citrus. Also a wide variety of whole grains, nuts, seeds and legumes; including chickpeas, red beans and lentils. Drink fruit juices or eat fresh fruit in the morning, vegetables the rest of the day. I would drink 32 to 64 ozs. of fresh organic vegetable juices each day (Reference books #3 & 4) also green drinksor green supplements. Use protein shakes made with whey or brown rice mixed in water, juice, rice or almond milk. Mixing brown rice with beans or with grains creates perfect proteins as well.

Millet cereal with brown rice is excellent for breakfast and is a natural source of IP-6. Use honey or black strap molasses as a natural sweetener. Eat ten raw almonds a day. Eat onions and garlic or take garlic supplements. As cancer cells break down, they produce ammonia. The amino acid arginine neutralizes ammonia. Arginine can be found in oats, soybeans, walnuts, wheat and wheat germ, in gelatin and collagen and in supplement form. Avoid exposure to chemicals (including treated water) and wash all fruits, vegetables and sprouts thoroughly. Try to eat raw food (uncooked) everyday. Use glass or stainless steel cookware. Avoid foods and beverages stored in aluminum. DO NOT USE ALUMINUM COOKWARE.

I would seek natural products to help support and detoxify my body. Options for detoxifying include: juice fasting, chelation therapy, colonics and supplements that support cleansing. To aid my body in cleansing I would take a colon cleanse and a parasite cleanse to free my body of accumulated toxins, parasites and other waste material. I would also cleanse the body of candida albicans, the microbes that are responsible for systemic yeast infections (all are discussed in reference book # 7).

Other supplements I would take include: plant-based digestive enzymes, probiotic intestinal micro-flora, whole leaf, cold processed aloe vera juice, colloidal minerals and antioxidants. Enzymes are involved in every process of the body including food digestion, building muscle, nerves, blood and bones. In fact, life could not exist without them. I would rebuild intestinal micro-flora with a probiotic supplement to increase absorption of vitamins, minerals, enzymes and fatty acids, support healthy bowel function, promote immunity and balance my body chemistry. Whole leaf, cold processed aloe vera juice will balance and feed the body, supports digestion, aid nutrient assimilation and has anti tumor properties. Colloidal minerals are readily absorbed; they promote vitality, support cellular repair, maintain and promote health. Antioxidants aid in repairing damaged tissue and protect us from free radical damage.

London, March 2 (ANI): Scientists have reached a step closer to developing potential treatments for devastating diseases including spinal cord injury, macular degeneration, diabetes and Parkinson’s disease, thanks to a new method of creating stem cells discovered by researchers at Mount Sinai Hospital in Canada.

The researchers say that their study accelerates stem cell technology, and provides a road map for new clinical approaches to regenerative medicine.

“We hope that these stem cells will form the basis for treatment for many diseases and conditions that are currently considered incurable,” Nature magazine quoted Dr. Andras Nagy, Senior Investigator at the Samuel Lunenfeld Research Institute of Mount Sinai Hospital, Investigator at the McEwen Centre for Regenerative Medicine, and Canada Research Chair in Stem Cells and Regeneration, as saying.

“This new method of generating stem cells does not require embryos as starting points and could be used to generate cells from many adult tissues such as a patient’s own skin cells,” the researcher added.

Dr. Nagy revealed that his method helps create pluripotent stem cells-which can develop into most other cell types-without disrupting healthy genes. The researcher added that the method involves a novel wrapping procedure to deliver specific genes to reprogram cells into stem cells.

Scientists have to date relied upon approaches that requite the use of viruses to deliver the required genes, a method that carries the risk of damaging the DNA.

Given that Dr. Nagy’s method does not require viruses, it overcomes a major hurdle for the future of safe, personalized stem cell therapies in humans.

“This research is a huge step forward on the path to new stem cell-based therapies and indicates that researchers at the Lunenfeld are at the leading edge of regenerative medicine,” said Dr. Jim Woodgett, Director of Research for the Samuel Lunenfeld Research Institute of Mount Sinai Hospital.

Regenerative medicine refers to enabling the human body to repair, replace, restore and regenerate its own damaged or diseased cells, tissues and organs. (ANI)

Effect of dietary chlorophyll derivatives on mutagenesis and tumor cell growth

Much attention in recent years has been given to the antigenotoxicity of chlorophyll. Chlorophyll, however, is known to be converted into pheophytin, pyropheophytin, and pheophorbide in processed vegetable food and following ingestion by humans. Studies were conducted on the antimutagenic and tumoricidal potencies of these compounds. All the chlorophyll derivatives tested exhibit identical antimutagenic effect towards 3-methylcholanthrene (3-MC), suggesting that the porphyrin nucleus may complex directly with the mutagen. It does not exclude, however, another mechanism of activity involving inactivation the enzymatic transformation of 3-MC. In contrast, the action of N’-nitro-N’-nitrosoguanidine (MNNG) depends upon structural differences between the chlorophyll derivatives. It is significantly lower when the phytol-containing pheophytin and pyropheophytin are tested as to that of the phytol-lacking pheophorbide. The higher concentrations of the chlorophyll derivatives were required to reduce the mutagenicity of MNNG than needed for 3-MC. The cytotoxicity of chlorophyll derivatives against tumor cells also was evaluated. The cellular uptake and inhibition of myeloma cell multiplicity were found to be greater for pheophorbide than for pheophytin. Calculated on the amount of cell associated chlorophyll derivative, however, pheophytin was more cytostatic/cytotoxic than pheophorbide. The results presented in this report indicate that food sources that yield chlorophyll derivatives may play a significant role in cancer prevention.

An Exciting New Discovery!

Recently the first stage of an extensive research project carried out at the Royal Victoria Hospital in Montreal, Canada produced some remarkable results. The project studies the effect of Aphanizomenon flos-aquae on the immune and endocrine systems, as well as on general blood physiology. It was discovered that eating AFA had a profound and unique effect on Natural Killer (NK) cells. The results were recently published under the title: Effects of the Blue Green Algae Aphanizomenon flos-aquae on Human Natural Killer Cells. It appears in Chapter 3.1 of the IBC Library Series, Volume 1911, Phytoceuticals: Examining the health benefit and pharmaceutical properties of natural antioxidants and phytochemicals.

NK cells have the ability to search for and recognize cells that are cancerous or have been infected by a virus, and kill them. The team of research scientists at the Royal Victoria Hospital, led by Dr. Gitte S. Jensen, discovered that eating Aphanizomenon flos-aquae triggers the movement of 40% of the circulating NK cells from the blood to the tissues where their main function is to perform immune surveillance and eliminate cancerous and virally-infected cells. Further research may prove that eating a small amount of AFA every day could assist in the prevention of cancer and viral infections. No other substance is known to trigger such a movement of NK cells in the body. This is very exciting news!
